A structure study of high-voltage cathode materials for lithium ion battery using X-ray crystallography

This diploma thesis deals with the X-Ray crystallography study of structural changes within lithium-ion cells. First part consists of the theoretical discussion about the structural changes in commerce lithium-ion cells with focus on X-Ray crystallography. Then the ex situ, in situ and operando terms are discussed. Verification of the XRD usage is realized on the LiFePO4 material. Experimental part deals with the optimization and preparation of electrochemical cells and the measurements. Finally, results of the Rietveld analysis are discussed.
Salts of 2-aminoethanol and ferrocene-based acids

Combination of the compact but sterically flexible ferrocene scaffold with intermolecular binding potential and conformational variability of (2-hydroxyethyl)ammonium structural motif was utilised in the construction of solid crystalline materials. Crystallisation in systems containing ferrocenecarboxylic, 2-ferrocenylacetic, 3-ferrocenylpropionic, 3-ferrocenyl- acrylic, 3-ferrocenylacrylic, 3-ferrocenylpropiolic, ferrocene-1,1'-dicarboxylic or ferrocene- sulfonic acid with 2-aminoethanol afforded crystals of the corresponding salts. The resulting crystalline products were characterised with the usual methods (proton nuclear magnetic resonance, infrared spectroscopy, elemental analysis) and their crystal structures were determined by means of single-crystal X-ray diffraction analysis. The crystal structures of the salts mentioned above usually contain rather complicated two dimensional networks of charge-assisted hydrogen bonds. With the aim of studying potentially more simple hydrogen-bonded structures, additional crystallisation experiments were conducted in systems of ferrocenecarboxylic acid and 2-(methylamino)ethanol or 2-(dimethylamino)ethanol. Complexes of metals as potential anion receptors

An important class of anion receptors are based on bimetallic complexes with phenols bearing an pendant arms in 2,6-positions. A ligand of this type, 2,6-bis[(N- methylpiperazine-1-yl)methyl]-4-formyl phenol (L1), was prepared using the Mannich reaction. An attempt to prepare a similar compound L2 yield only an oily product. The molecular structure of compound L1 was determinated by X-ray diffraction. Comparison of the L1 structure with similar ones from CCDC showed important influence of the intramolecular H-bond on pendant arms orientations in this type of compounds. Further synthetic exploitation of the aldehyde group (in the position 4) was tested by reaction with 4-nitroaniline. The Shiff base (L3) was obtained but only as an oily substance. Reactions of L1 with Cu(ii) and Zn(ii) perchlorate or acetate produce complexes; the compound [Cu2L1(OH)(OAc)(H2O)]ClO4 (K1) was obtained in a crystallic form. The interactions of anion-bimetallic complex receptor were discused from stereochemical point of view using the structural data from CCDC in the last part of this work.
Preparation and study of human lymphocyte receptor LLT1

Natural killer (NK) cells are an intensively studied part of immune system, possessing unique ability to recognize and induce death of tumor and virus-infected cells without prior antigen sensitization. Their function is regulated by a fine balance of signals induced by multiple activating and inhibitory cell surface receptors and their interaction with the ligands present on the target cell. Recent research in their C-type lectin-like receptors repertoire has shown that ligands of some of these previously orphan receptors lie within their own family, describing a lectin-lectin interaction. This is the case of human inhibitory receptor NKRP1 (gene KLRB1) and its ligand LLT1 (gene CLEC2D). Previous studies have shown that overproduction of LLT1 in cancer cells or lower production of NKRP1 in NK cells is connected to cancerous manifestations. This master's thesis shows a successful production of the extracellular part of LLT1 utilizing a mammalian expression system based on transient transfection of modified human embryonic kidney (HEK) cell lines. It was found that the five cystein residues contained within the lectin domain of LLT1 tend to cause misfolding and formation of aggregates. Stabilization of the domain was achieved by restoration of the sixth cystein residue at the evolutionary conserved...
Alternative synthesis of of 1-(diphenylphosphino)-1'-(N,N-dimethylaminomethyl)ferrocene

An alternative, more straightforward and effective method for the preparation of 1- (diphenylphosphino)-1'-(N,N-dimethylaminomethyl)ferrocene was developed using 1-bromo- 1'-(diphenylphosphino)ferrocene as the starting material and Eschenmoser's salt as an aminomethylation agent. Purity of thus prepared phosphinoamine was verified by 1 H and 31 P NMR spectroscopy and by comparison of the data with those reported in the literature (M. E. Wright, Organometallics, 1990, 9, 853). The coordination chemistry of this compound was investigated in gold(I) complexes. Thus, chlorido[1-(diphenylphosphino)-1'-(N,N-dimethyl- aminomethyl)ferrocene]gold(I) complex was synthesized and converted to ill-defined [1- (diphenylphosphino)-1'-(N,N-dimethylaminomethyl)ferrocene]gold(I) perchlorate by halogen abstraction with AgClO4. Protonization of the nitrogen atom in both compounds with hydrogen chloride yielded the corresponding hydrochlorides. It was shown that 1- (diphenylphosphino)-1'-(N,N-dimethylaminomethyl)ferrocene is coordinated as a simple phosphine in the resulting complexes while its amine nitrogen is protonated. All these complexes were characterized by means of 1 H, 31 P and 13 C NMR spectroscopy, mass spectroscopy with electrospray ionisation, elemental analysis and by single-crystal X-ray diffraction analysis.

Structure Analysis of Plant Bifunctional Nuclease TBN1

This work is dedicated to thorough structural analysis of plant bifunctional nuclease TBN1, the representative of plant nuclease I group. TBN1 along with homologous nucleases from this family plays an important role in plant cell life cycle and also shows considerable anticancerogenic effects. Two variants of TBN1 (wild type and N211D mutant) were studied. Properties of both variants in different solutions were analyzed. Both were successfully crystallized. Structures of both types of TBN1 were solved using X-ray diffraction. The phase problem was solved by Multi-wavelength anomalous dispersion using Zn2+ ions natively present in TBN1. Structural properties of TBN1 such as fold, active site composition, effect of glycosylation and surface electrostatic potential distribution were analyzed. Reaction mechanism of TBN1 was proposed on the basis of structural properties and comparisons with similar structures.
